Se necesita una forma romboidal (plantilla) como las que se muestran a continuación, que puede ser de plástico o metal u otros materiales (véase la figura de al lado). Sus dimensiones deben ser tales que cubra 10×10= 100 células. Considerando que existen en el comercio planchas de cera con diferentes tamaños de celdas y, sobre todo, que éstas pueden variar con los diversos ciclos de cría, puede decirse que aproximadamente el rombo debe tener cada uno de los lados 5,4-5,5 cm; el lado oblicuo debe tener una inclinación de 60°. Cabe señalar que, a efectos comerciales, aunque el rombo fuera unos mm más pequeño o más grande no supondría un gran problema, siempre y cuando se utilice siempre la misma plantilla para las mediciones entre las distintas familias.Como alternativa a la plantilla, se puede marcar con un rotulador la primera celda de la esquina superior izquierda y, a partir de ahí, contar 10 celdas a la derecha; después se cuentan 10 celdas en diagonal y, a continuación, se marca la celda de la esquina inferior derecha (véase la figura de al lado). De este modo, se puede circunscribir el perímetro del rombo de 10 x 10 y, a continuación, perforarlo. Un alfiler preferiblemente de diámetro bastante grande (1-1,5 mm). Pasos del procedimiento Seleccionar y marcar un panal de cría lo más compacto posible. Prefiera los panales centrales del nido para reducir los posibles efectos de las diferentes temperaturas de la cría. En el panal seleccionado, buscar una zona que tienda a ser central, en la que no haya muchas celdas vacías (estas últimas no deben superar el 10%) y colocar la forma romboidal en esta zona. Marque las esquinas de la plantilla con un rotulador (por ejemplo, UniPosca blanco) para poder identificar la zona que debe comprobarse los días siguientes. Como alternativa, si la plantilla tiene un borde curvado que se apoya directamente sobre la cría, se puede aplicar una ligera presión para que todo el perímetro de la plantilla quede impreso sobre la cría. Tome nota del número de celdas ya vacías dentro de la plantilla. Proceder a perforar todas las celdillas operculadas dentro de la plantilla. Volver a colocar el panal dentro del nido (preferentemente en la misma posición inicial). Vuelva al cabo de 24 h y cuente las células limpias (vacías) en la zona previamente marcada. Efectuar los cálculos mediante la fórmula siguiente Los valores superiores al 90 % se consideran buenos. Valores inferiores al 70 % pueden ser indicativos para considerar la sustitución de la reina. Para una mayor fiabilidad es aconsejable repetir el test dos veces durante la temporada, evitando los periodos de fuerte importación (los mejores periodos pueden ser al principio y al final de la temporada).
